Inthisimproved,rapidmethod,theincubationtimewithPAA+AMGisreducedto4handthelevelsofbothPAAandAMGareincreasedtoensurethatresistantstarchlevelsobtainedwithasetofcontrolsamplesareconsistentwithileostomydata.Undertheseconditions,theDFvaluesobtainedformostsamplesarethesameasthoseobtainedwithAOACMethods2009.01and2011.25.
Thedietaryfiberfractionsthataremeasuredwiththismethodare:
1.HighMolecularWeightDietaryFiber(HMWDF)includingInsolubleDietaryFiber(IDF)andHigh MolecularWeightSolubleDietaryFiber(SDFP;solubledietaryfiberwhichisprecipitatedinthepresenceof78%aqueousethanol),and
2.LowMolecularWeightSolubleDietaryFiber(SDFS;watersolubledietaryfiberthatissolubleinthepresenceof78%aqueousethanol).
Alternatively,IDF,SDFPandSDFScanbemeasuredseparately.
Theenzymesusedinthismethodarehighpurityandeffectivelydevoidofcontaminatingenzymesactiveonotherdietaryfibercomponentssuchas
