Background:oralcancerisaresultofdisorderedcellularbehaviorinitiatedbyvariousstimuliwhichischaracterizedbythealterationofserumglycoproteinsconsistingofdifferentmonosaccharides.Oneoftheseislevo-fucose(L-fucose),amethylpentose.Elevatedlevelsofprotein-boundfucosehavebeenreportedinvariousmalignancies.Aim:ThepresentstudyattemptedtocorrelatelevelsofserumL-fucoseasabiomarkerwiththevarioustumornodemetastasis(TNM)stagesoforalcancer.Methodology:Thestudywascarriedouton90subjectsconsistingof30healthycontrolsand60histopathologicallyprovenoralsquamouscellcarcinoma(OSCC)cases.TheserumfucoselevelestimationwasdonebasedonthemethodadoptedbyWinzler.Statisticalanalysisincludedindependentsample"st-test,one-wayANOVAtest,Karl–Pearsoncorrelationtest,andTukey"sHSDposthoctesttoevaluatethesignificanceandvariABIlityofvaluesbetweengroups.Results:SignificantelevationinserumfucoselevelswasnoticedamongOSCCpatientswhencomparedwiththecontrolsandaprogressiveascentofL-fucoselevelswerenotedasthestageofseverityincreased.SerumfucoselevelswereindependentofhistopathologicalgrADIng,age,andsex.Conclusion:SerumL-fucoselevelswereincreasedinOSCCpatients,andapositivecorrelationwasobservedbetweenserumL-fucoselevelsandTNMstagingofOSCC.Thus,serumL-fucosecanbeusedasaneffectivediagnosticandprognosticbiomarkerinOSCCpatients.

TheperiodontalpathogenTannerellaforsythiaexpressesseveralglycosidaseswhicharelinkedtospecificgrowthrequirementsandareinvolvedintheinvasionofhosttissues.α-L-Fucosylresiduesareexposedonvarioushostglycoconjugatesand,thus,theα-L-fucosidasespredictedintheT.forsythiaATCC43037genomecouldpotentiallyserverolesinhost-pathogeninteractions.WedescribethemolecularcloningandcharacterizationoftheputativefucosidaseTfFuc1(encodedbythebfo_2737=Tffuc1gene),previouslyreportedtobepresentinanoutermembranepreparation.Intermsofsequence,this51-kDaproteinisamemberoftheglycosylhydrolasefamilyGH29.Usinganartificialsubstrate,p-nitrophenyl-α-fucose(KM 670 µM),theenzymewasdeterminedtohaveapHoptimumof9.0andtobecompetitivelyinhibitedbyfucoseanddeoxyfuconojirimycin.TfFuc1wasshownheretobeauniqueα(1,2)-fucosidasethatalsopossessesα(1,6)specificityonsmallunbranchedsubstrates.Itisactiveonmucinaftersialidase-catalyzedremovalofterminalsialicacidresiduesandalsoremovesfucosefrombloodgroupH.Followingknock-outoftheTffuc1geneandanalyzingbiofilmformationandcellinvasion/adhesionofthemutantincomparisontothewild-type,itismostlikelythattheenzymedoesnotactextracellularly.BiochemicallyinterestingasthefirstfucosidaseinT.forsythiatobecharacterized,thebiologicalroleofTfFuc1maywellbeinthemetabolismofshortoligosaccharidesintheperiplasm,therebyindirectlycontributingtothevirulenceofthisorganism.TfFuc1isthefirstglycosylhydrolaseintheGH29familyreportedtobeaspecificα(1,2)-fucosidase.

α-L-Fucosidasesarekeyenzymesforthedegradationofintestinalglycansbygutmicrobes.Inthiswork,threeputativeα-L-fucosidases(Afc1,Afc2,andAfc3)genesfromClostridiumperfringensATCC13124wereclonedandexpressedinEscherichiacoli.Afc1hadtheα-L-fucosidasedomainofglycosidehydrolase(GH)29familybutshowednoenzymeactivitytowardallthesubstratesexamined.Theputativeacid/baseresidueofAfc1,Ser205,wasreplacedbyaglutamicacidwhichisconservedinGH29-Bα-L-fucosidases.However,themutantAfc1-S205Estillfailedtoshowenzymeactivity.Afc2andAfc3weredeterminedtobe1,3-1,4-α-L-fucosidaseofGH29-Bsubfamilyand1,2-α-L-fucosidaseofGH95family,respectively,andbothofthemcouldreleasefucosefromporcinegastricmucin(PGM).WhenC.perfringensATCC13124grewwiththepresenceofPGM,thetranscriptionofafc1decreasedslightly,whilethoseofafc2andafc3increasedto2.2-foldand1.4-fold,respectively,andtheenzymeactivitiesofAfc2andAfc3inthecultureincreasedto2.2-foldand2.6-fold,respectively.TheseresultssuggestthatAfc2andAfc3areinvolvedinthedegradationofintestinalfucosylglycansbyC.perfringensATCC13124.
