RiparianhABItatmakesupasmallfractionofthelandscapebutprovidesimportantstopoverhabitatformigratorybirds.Hydroelectricdamoperationscausefluctuationsinwaterlevelsthatcanchangetheamountorqualityofriparianhabitat,whichinturnmightaffectpotentialfatteningratesofmigrantbirds.Hereweusedplasmametaboliteanalysistoestimatevariationinfatteningrateinrelationshiptovariablewaterlevelsassociatedwithreservoirmanagementinfourspeciesofneotropicalmigratorysongbirdsusingriparianhabitatatadam-impactedstopoversiteinRevelstoke,BritishColumbia,Canada.Residualplasmatriglyceride,ourmeasureofestimatedfatteningrate,variedsystematicallywithtimeofdayandJuliandateandvariedconsistentlyamongspecies,butdidnotvarywithageorsex.Controllingforpotentiallyconfoundingvariables,wefoundnointer-annualvariationinestimatedfatteningrate,eventhoughthereweremarkeddifferencesinwaterlevelsamongyears.Likewise,therewasnorelationshipbetweendailyvariationinwaterlevelsandestimatedfatteningrate.Dataonfeatherisotopes(δD),indicativeofmigratoryorigin,didnotaddexplanatorypowertoourmodels.Therewasinter-annualvariationinplasmaglycerolandβ-hydroxybutyratelevelsandsignificant,thoughweak,relationshipsbetweenthesemetabolitesandwaterlevel(highermetabolitelevelswhendrier)thatmightindicateeffectson‘bodycondition’independentoffatteningrate.Ourstudysuggeststhat,atpresent,althoughhydroelectricdamoperationsinfluencewaterlevelsintheArrowsLakeReservoirandadjacentriparianhabitats,thisdoesnotsignificantlyimpactfatteningratesofmigratorypasserinesusingthesehabitats.

Background:Acetateisacommonmicrobialfermentativeend-product,whichcanpotentiallybeusedasasupplementarycarbonsourcetoenhancetheoutputofbiotechnologicalproductionsystems.ThisstudyfocusesontheacetatemetabolismofthephotosyntheticcyanobacteriumSynechocystissp.PCC6803whichisunabletogrowonacetateasasolecarbonsourcebutstillcanassimilateitviaacetyl-CoA—derivedmetabolicintermediates.Inordertogaininsightintotheacetateuptake,associatedlimitationsandmetaboliceffects,aheterologousacetatetransporterActPfromEscherichiacoliwasintroducedintoSynechocystistofacilitatethetransportofsupplementedacetatefromthemediumintothecell.Results:Theresultsshowthatenhancedacetateintakecanefficientlypromotethegrowthofthecyanobacterialhost.Theeffectisapparentspecificallyunderlow-lightconditionswhenthephotosyntheticactivityislow,andexpectedtoresultfromincreasedavailabilityofacetyl-CoAprecursors,accompaniedbychangesinducedincellularglycogenmetabolismwhichmayincludeallocationofresourcestowardsenhancedgrowthinsteadofglycogenaccumulation.Despitethestimulatedgrowthofthemutant,acetateisshowntosuppresstheactivityofthephotosyntheticapparatus,furtheremphasizingthecontributionofglycolyticmetabolismintheacetate-inducedeffect.Conclusions:TheuseofacetatebythecyanobacteriumSynechocystissp.PCC6803isatleastpartiallyrestrictedbytheimportintothecell.Thiscanbeimprovedbytheintroductionofaheterologousacetatetransporterintothesystem,therebyprovidingapotentialadvantagebyexpandingthescopeofacetateutilizationforvariousbiosyntheticprocesses.
